钙调蛋白磷酸酶Calcineurin对构巢曲霉产孢功能的影响

         

摘要

Asexual sporulation is a common reproductive mode for a diverse group of medical fungi. Aspergillus nidulans,a premier model fungus for filamentous fungi,is used in this study.On the base of successfully constructed theΔcnaA andΔcnaB strains,we investigate the biological function of Calci-neurin in the regulation of asexual sporulation from the following three aspects:① Comparison of conidiation phenotypes between calcineurin deletion mutants and wild type strain;② Comparison of quantification conidia between calcineurin deletion mutants and wild type strain;③ Characterization of conidiophores defects in calcineurin mutants indicated by the scanning electron micrographs (SEM).Our results suggest that Calcineurin,especially its regulatory subunit B (cnaB),is critical for conidial devel-opment,with the decreased number of conidiospores and abnormal phialides in calcineurin mutants. Nevertheless,our findings in the present study will provide the theoretical basis for us to control patho-genic microorganism and be of great help in finding novel drug therapies for fungal infections.%无性产孢是医学上多种丝状真菌最为普遍的繁殖方式。以模式生物构巢曲霉为研究对象,通过观察钙调蛋白磷酸酶Calcineurin敲除菌株的菌落表型、产孢量的统计,以及扫描电镜分析产孢结构及其分生孢子的表面结构特征,探讨Calcineurin在构巢曲霉产孢过程中的功能。结果表明,Calcineurin尤其是其调节亚基 CnaB严重影响构巢曲霉的产孢,引起产孢结构和分生孢子的异常。此结果将有助于筛选出新的真菌特异性药靶,为真菌感染防治提供新策略。
