丹龙醒脑方对局灶性脑缺血大鼠CA1区Nestin和GFAP表达的影响

         

摘要

Objective Explore the influence of Danlongxingnao recipe on expression of Nestin protein and GFAP protein in rats CA1 area after focal cerebral ischem ia/reperfusion. Methods The 48 rats w ere randomly divided into sham-operation group, modelling group, Danlongxingnao recipe group and edaravone group.T he m odle of cerebral ischem ia reperfusion (I/R )was induced by middle cerebral artery occlusion (MCAO) and recanalization. Neurological sym ptom score were evaluated. HE staining of the cells was used to observe the morphology changes in hippocam pus CA1 area. Nestin protein and GFAP Protein were observed by im munohistochemical method in CA1 area. Results Compared with sham-operation, neurological symptom score of model group were increased and the result was significant (P<0.05) after 3d and 7d treatm ent. Compared with model group, neurological symptom score of Danlongxingnao recipe group and edar-avone group were decreased and the result was significant (P<0.05) after 3d and 7d treatm ent. The expression of average optital density of Nestin protein in Danlongxingnao recipe increased after 3d and 7d treatment (P<0.05) and the expressions of average gradation of GFAP protein in Danlongx-ingnao recipe decreased after 3d and 7d treatment (P<0.05). Conclusion The Danlongxingnao recipe had protective effects on the pathologicalm orphological changing in hippocam pus CA1 area. Danlongxingnao recipe can make effects after focal cerebral ischem ia/reperfusion and it may be as-sociated with promoting the proliferation and differentiation of neural stem cells.%目的:探讨丹龙醒脑方对大鼠脑缺血损伤后海马CA1区巢蛋白(Nestin)和胶质纤维酸性蛋白(glial fib-rillary acidic protein,GFAP)表达的影响。方法将48只大鼠随机分为假手术组、模型组、依达拉奉组、丹龙醒脑方组。采用大脑中动脉线栓法制备大脑中动脉闭塞再灌注(MCAO)模型,对大鼠进行神经功能缺损评分,采用HE染色观察海马CA1区细胞的形态学改变,采用免疫组化法检测海马CA1区Nestin 蛋白和GFAP蛋白的表达。结果与假手术组比较,模型组治疗3 d后和7 d后的神经功能缺损评分明显升高,差异有统计学意义(P<0.05);与模型组比较,依达拉奉组和丹龙醒脑方组治疗3 d后和7 d后的神经功能缺损评分明显降低,差异有统计学意义(P<0.05);丹龙醒脑方组在治疗3 d和7 d后Nestin蛋白的平均光密度增加(P<0.05),GFAP蛋白的平均灰度值减少(P<0.05)。结论丹龙醒脑方对海马CA1区病理形态有保护作用。丹龙醒脑方抗脑缺血损伤可能与促进海马区神经细胞增殖和分化有关。
