丝素蛋白和丝胶蛋白中氨基酸的测定

摘要

以经济昆虫家蚕(Bombyx mori L.)为研究对象，对家蚕(Bombyx mori L.)蚕茧中提取分离的丝素蛋白和丝胶蛋白，采用考马斯亮蓝法分别测定两种蛋白含量，然后通过薄层层析法分离鉴定两种蛋白中氨基酸种类。结果表明丝素蛋白和丝胶蛋白中均含有甘氨酸、丙氨酸和精氨酸。进一步用茚三酮法测定脯氨酸含量，为分析丝素蛋白和丝胶蛋白中氨基酸组成和含量奠定基础。%Bombyx mori L.is a kind of economic insect as the research object, silk fibroin and sericin protein are extracted from from silkworm cocoon, and contents of two proteins are detected by coomassie brilliant blue G250 dye ,then the types of amino acid are identified by thin-layer chromatography method. The result shows the two proteins consist of Glycine, alanine and arginine. On the basis of this study, the content of proline is measured by ninhydrin reaction. The research establish a basis on detecting component and content of silk fibroin and sericin protei n.
