采用TTC(2,3,5-氯化三苯基四氮唑)法测定花粉活力,研究了温度、相对湿度、蔗糖含量、硼酸含量等因子对野生黄连花粉活力的影响,以期为探索梵净山野生黄连的生殖机制提供理论依据.结果表明,当温度为26℃、相对湿度为60%时,梵净山野生黄连花粉活力达最大值(45.9%),比温度20℃、相对湿度70%时的花粉活力(10.5%)增加35.4个百分点.随着温度和相对湿度的增加或降低,梵净山野生黄连花粉活力均呈下降的趋势.当蔗糖含量20%、硼酸含量0.006%时,花粉活力为93.4%,当蔗糖含量大于或小于20%,硼酸含量大于或小于0.006%时,梵净山野生黄连的花粉活力均表现为下降.因此,在梵净山野生黄连开花时,控制温度26℃、相对湿度60%,同时喷施20%蔗糖、0.006%硼酸,可以大大提高黄连的花粉活力.%In order to find key factors affecting pollen vitality,and to provide a theoretical basis for exploring the reproductive mechanism of Coptis chinensis Franch. in Fanjingshan mountain area, the influence of temperature, relative humidity, saccharose and element B on pollen vitality of Coptis chinensis Franch. were studied, using TTC (2, 3, 5-triphenyl tetrazolium chloride) test. The results showed when the temperature was 26 ℃, relative humidity was 60%, pollen vitality reached the maximum (45. 9%). Under this condition, pollen vitality was 35. 4 percentage points higher than that at 20 ℃ with relative humidity of 70%(10. 5%). Pollen vitality of Coptis chinensis Franch. was at a tendency of declining with the temperature and relative humidity increased or decreased. When the concentration of saccharose was 20% and the concentration of H3BO3 was 0. 006% ,pollen vitality was 93. 4% ,and it showed a tendency of declining with the concentration of saccharose and H3BO3 increased or decreased. Under temperature of about 26 ℃ and relative humidity of about 60%, with 20% saccharose and 0. 006% H3BO3 as well,po!2en viability of Cop-its chinensis Franch. can be boosted greatly.
