苦参碱和氧化苦参碱对体外培养兔视网膜色素上皮细胞增殖的影响

     

摘要

目的 观察苦参碱和氧化苦参碱对体外培养兔视网膜色素上皮细胞(retinal pigment epithelial cell,RPEC)增殖的影响,并与苏拉明进行比较.方法 生长状态良好的第4代RPEC接种于3块96孔细胞培养板,每块培养板上设苦参碱、氧化苦参碱、苏拉明各5个浓度组及空白对照组各5个复孔.培养24、48、72h时各取1块培养板,噻唑蓝比色法测定每孔的吸光度值,计算细胞增殖抑制率,对各个药物浓度组在不同作用时间的吸光度值与空白对照组进行统计学比较,并求得3种药物在加药培养72h时的半数抑制率(50% inhibiting concentration,IC50)浓度.结果 苦参碱、氧化苦参碱和苏拉明对RPEC的增殖均有抑制作用,随着药物剂量的增加和作用时间的延长,3种药物对细胞增殖的抑制率都逐渐增加.苦参碱、氧化苦参碱和苏拉明作用72h的IC50浓度分别为74.89、141.61和186.24mg/L.结论 苦参碱和氧化苦参碱对体外培养兔RPEC的增殖均有明显的抑制作用,呈现时间和剂量依赖性,苦参碱作用较强.%Objective To investigate the effect of matrine and oxymatrine on the proliferation of rabbit retinal pigment epithelial cells (RPEC) cultured in vitro , and to compare with suramin. Methods The 4th passage lPEC in good condition were plated in three 96 -well culture plates. In each plate,there were 5 wells for each of 5 different concentrations of matrine, oxymatrine and suramin and for the control group. All plates were used for the mensuration of absorption value by methyl thiazolyl tetrazolium colorimetric method after 24, 48 or 72h culture, respectively. The inhibitory rate of each drug concentration at each time point and the IC50 concentration of each drug after 72h culture were analyzed.Statistical analysis between the absorption values of each drug concentration group and the control group at each time point were completed. Results The inhibitory effect on the proliferation of lPEC was found in all the matrine, oxymatrine and suramin group, and the inhibitory rates of all drugs increased gradually with the increasing of drug dose and action time. After 72h cultue, the IC50 concentrations of matrine,oxymatrine and suramin were 74.89,141.61and 186.24mg/L, respectively. Conclusion There are significant inhibitory effects of matrine and oxymatrine on the proliferation of rabbit RPEC cultured in vitro in time -dependent and dose -dependent manners. Matrine is comparatively stronger.
