目的 研究受体酪氨酸激酶Axl对肝癌细胞生长的影响及其相关的作用机制.方法 体外培养肝癌细胞(hepatom a carcinoma cell,HepG2),用小干扰RNA(small interfering RNA,siRNA)干扰HepG2细胞的Axl表达,生长曲线法检测干扰前后HepG2细胞生长情况;Western blot方法检测生长信号通路中Akt、p-Akt的表达水平.结果 HepG2细胞的Axl经siRNA干扰表达下降后细胞生长受到抑制;Western blot结果显示,干扰后细胞增殖信号通路中主要蛋白p-Akt的表达明显降低.结论 Axl可能通过Akt信号通路影响肝癌细胞的生长.%Objective To investigate the functional role of Axl receptor tyrosine kinase on the growth of hepato - carcinoma cells and the underlying molecular mechanisms. Methods Small interfering RNA( siRNA) were used to silence the expression of the Axl on the cultured hepatom a carcinoma cell( HepG2) cells. The effects of the Axl receptors on cell survival were tested by cell growth experiments. The expression and activation of Akt, a key signaling molecule in the proliferation - related signaling transduction pathway were analyzed by Western blot. Results Inhibition of Axl expression by siRNA significantly delayed the HepG2 cell growth , in consistent with a dramatic down - regulation of the Akt activation. Conclusion Axl regulates the growth of hepatocyte carcinoma cells through Akt signaling pathway.
