NUCLEAR-TRANSFER SIGNAL TO INTRODUCE HUMAN HEPATIC CARCINOMA CELL-DERIVED GROWTH FACTOR INTO NUCLEUS, AND NUCLEAR-TRANSFER SIGNAL TO INTRODUCE MOUSE HEPATIC CARCINOMA CELL-DERIVED GROWTH FACTOR INTO NUCLEUS
PROBLEM TO BE SOLVED: To identify the subject nuclear-transfer signal useful for introducing proteins having a various kind of physiological activity. ;SOLUTION: This signal peptide is a nuclear-transfer-signal peptide (hHDGF-NLS1) for human hepatic carcinoma cell-derived growth factor (hHDGF), having an amino acid sequence of formula I, or a nuclear-transfer- signal peptide (hHDGF-NLS2) for human hepatic carcinoma cell-derived growth factor (hHDGF), having an amino acid sequence of formula II. It is identified where a nuclear-transfer signal is on hHDGF, by deleting a part of the hHDGF gene, preparing a variety of hHDGF variants, constructing an expression plasmid of a complex gene product with these and β-galactosidase normally absent in the nucleus, and including this in a nucleated cell. Then two nuclear-transfer signals can be found by homology-screening of the amino acid sequence of hHDGF over the data base.;COPYRIGHT: (C)1998,JPO
