小G蛋白Rab40a的亚细胞定位及功能研究

         

摘要

Objective To study the subcellular localization and intracellular vesicle transport pathway which small GTPase Rab40a is involved in. Methods The colocalization of green fluorescent protein ( GFP) labeled Rab40a with subcellular structures, especially transport vesicles in Hela cells was observed by confocal microscopy. The intracellular vesicular transport pathways which Rab40a may be involved in was studied by using RNA silencing technology. Results Rab40a was colocalized with endosome and Golgi marker proteins, EEA1 and Golgin-97. Interfering Rab40a blocked the transportation of cholera toxin B from endosome to Golgi. Conclusion Rab40a is localized in endosome and Golgi and participates intracellular transport from endosome to Golgi.%目的 研究小G蛋白Rab40a的亚细胞定位及参与的细胞内囊泡运输路径.方法 应用共聚焦显微镜技术,研究绿色荧光蛋白(green fluorescent protein,GFP)标记的Rab40a在Hela细胞内与各种亚细胞结构,特别是转运囊泡的共定位.利用RNA沉默技术研究Rab40a可能参与的细胞内囊泡转运过程.结果 Rab40a与高尔基标记蛋白Golgin-97、内吞体标记蛋白EEA1存在共定位;干扰Rab40a表达明显阻断了霍乱毒素B自内吞体向高尔基体的转运.结论 Rab40a定位于细胞内吞体和高尔基体,参与细胞内内吞体到高尔基体之间的转运.
