目的:探讨癌-睾丸抗原SSX-2基因在儿童急性淋巴细胞性白血病(ALL)外周血单个核细胞中的表达,为判断ALL预后提供理论依据.方法:采用逆转录-聚合酶链反应,对51例ALL患儿外周血单个核细胞进行SSX-2 mRNA检测,并进行SSX-2基因表达与临床特征的相关性分析;以30例非肿瘤患儿为对照组.结果:ALL初治患儿SSX-2 mRNA表达率为34.78%,对照组无一例表达.ALL初治组表达率明显高于对照组(P<0.05),ALL初治组表达率高于ALL完全缓解组表达率7.14%(P<0.05).SSX-2基因的表达与儿童ALL性别、年龄、初诊时外周血白细胞数、免疫分型以及临床危险分级无关(P>0.05).结论:儿童ALL患者SSX-2 mRNA表达率较高,SSX-2 mRNA表达可能与儿童ALL的发生、发展有关;SSX-2基因检测对儿童ALL近期疗效的评估、指导临床治疗有一定意义.%Objective:To investigate the expression of SSX-2 gene and its relationship and clinical implication in children with acute lymphoblastic leukemia( ALL). Methods:Reverse transcription polymerase chain reaction (RT-PCR)was used to detect SSX-2 mRNA expression in peripheral blood mononuclear cells of 51 children with ALL and 30 children with no-malignant diseases served as control group. Results:The rate of SSX-2 mRNA expression in children in newly diagnosed ALL was 34. 78%, while no case was positive in control group, which was remarkably higher in newly diagnosed ALL group than control group ( P < 0. 05). Especially, the positive expression rate of SSX-2 was 34. 78% in newly diagnosed ALL, whereas it was 7. 14% in complete remission in children with ALL( P <0. 05). The expression of SSX-2 gene was not associated with clinical characteristics (gender, age, WBC, immunophenotype, clinical risk) in children with ALL( P >0. 05). Conclusion: There is a high expression of SSX-2 mRNA in children with ALL. The expression of SSX-2 gene might play an important role in the pathogenesis of children with ALL. Examination of SSX-2 mRNA might have clinical significance guiding to select adaptive therapy in children with ALL.
