[Objective] To study the method for rapid identification of genetic sex in Hainan medaka (Oryzias curvinotus).[Method] DNA was extracted from Hainan medaka. Design of sex-specific primers Ocsex-F/Ocsex-R was based on genomic data. The genetic sex of the Hainan medaka was identified by PCR.[Result]The primers Ocsex-F/Ocsex-R were amplified in male DNA to obtain two DNA bands of 959 bp and 656 bp, but only a single band of 959 bp could be amplified in females. The same results appeared in Hainan medaka populations from Shantou, Zhanjiang, Qinzhou, Wenchang.[Conclusion]The primers Ocsex-F/Ocsex-R had stability and versatility in different populations of the South China Sea coast, which could be widely used for the genetic sex identification of Hainan medaka.%[目的]研究快速鉴定弓背青鳉遗传性别的方法.[方法]以中国南海沿岸弓背青鳉群体为对象提取基因组DNA,根据基因组数据设计性别特异引物Ocsex-F/Ocsex-R,通过PCR方法对弓背青鳉遗传性别进行鉴定.[结果]引物Ocsex-F/Ocsex-R在雄性DNA中扩增获得959 bp和656 bp的两个DNA条带,在雌鱼中仅可扩增出959 bp的单一条带,PCR产物经电泳可清晰分辨;对汕头、湛江、钦州、文昌等南海沿岸的地理群体扩增结果一致.[结论]Ocsex-F/Ocsex-R引物可对弓背青鳉进行遗传性别鉴定,在南海沿岸不同群体中具有稳定性和通用性,可广泛用于弓背青鳉遗传性别鉴定.
