目的 采用病理学及代谢组学方法研究PSGL-1缺失对巨噬细胞脂质吞噬能力及代谢组的影响.方法 提取小鼠原代腹腔巨噬细胞,进行油红O染色,采集样本的1 H NMR谱并运用多变量分析方法进行代谢特征的分析.结果 PSGL-1-/-;LDLR-/-小鼠的腹腔巨噬细胞中脂质面积减少;与LDLR-/-鼠比较,PSGL-1-/-;LDLR-/-小鼠巨噬细胞代谢物中牛磺酸和三甲胺的含量上调,而脂质、乳酸、丙氨酸和琥珀酸盐含量降低.结论 PSGL-1缺失影响腹腔巨噬细胞吞噬脂质的功能,而且能够调控LDLR-/-小鼠巨噬细胞的代谢状态.%Objective To study the effects of PSGL-1 deletion on macrophage lipid phagocytosis and metabolic profiles by the use of pathologcal and metabonomics methods. Methods Primary mouse peritoneal macrophages were extracted and stained with oil red O, and the 1 H NMR spectra of the samples were collected and analyzed by multivariate analysis. Results The lipid area in the peritoneal macrophages of PSGL-1-/ -; LDLR-/ - mice was decreased. Compared to LDLR-/ - mice, the peritoneal macrophages in PSGL-1-/ -; LDLR-/ - mice exhibited higher levels of taurine and trimethylamine, and lower levels of the lipid, lactate, alanine and succinate. Conclusion PSGL-1 deletion can affect the function of macrophage phagocytosis of lipids, and regulate the metabolic state of macrophages in LDLR-/ - mice.
