Objective:To select the most suitable macroporous resin for the purification of astragaloside in astragaloside extract from three kinds of macroporous resins,and optimize it by static and dynamic adsorption-desorption process.Methods:Static adsorption and desorption performance of three kinds of macroporous adsorption resins (D101,D201 and AB-8) for astragaloside Ⅳ from astragaloside extract were investigated to screen the op timal resin.The adsorption flow rate,sample pH value,adsorption time,elution solvent concentration and elution flow rate were tested by single factor experiment to enrich the content of astragaloside Ⅳ.The dynamic adsorption and desorption condition of resin were optimized,and the purity of astragaloside Ⅳ was evaluated in the end.Results:AB-8 macroporous resin was chosen as the optimal resin.As a result,adsorption pH of 5,adsorption time of 8 h,ethanol elution of 50%,adsorption rate of 1 BV/h and elution speed of 1 BV/h were derived as optimal conditions.Conclusion:The process about adsorption and desorption of astragaloside Ⅳ mentioned above was the best method.And it can get a higher content of astragaloside Ⅳ.%目的 从3种大孔树脂中筛选出最适合黄芪甲苷提取液中黄芪甲苷纯化的大孔树脂,并对其进行静态、动态吸附-解吸工艺条件优化.方法 以吸附及解吸附能力为指标,比较D101、D201及AB-8 3种大孔树脂对黄芪甲苷的吸附效率,通过单因素实验考察吸附流速、上样样品pH值、静态吸附时间、洗脱溶剂浓度及洗脱流速对提取黄芪甲苷含量的影响,优选树脂的动态吸附及解吸附条件,并评价纯化后得到黄芪甲苷的纯度.结果 黄芪甲苷采用AB-8大孔树脂进行吸附,最佳吸附pH值为5,最佳吸附速率为1 BV/h,静态吸附8h,采用50%的乙醇洗脱,最佳洗脱速度为1 BV/h.结论 上述所建立的大孔树脂纯化黄芪甲苷的吸附和解吸附效果最好,得到的黄芪甲苷含量较高.
