目的 观察芦荟多糖对炎症性肠病(IBD)小鼠免疫功能及对OX40、OX40L表达的影响并探讨其机制. 方法 采用三硝基苯磺酸(TNBS)诱导法制备IBD小鼠模型,随机分为模型组、柳氮磺胺吡啶(SASP)组(514 mq/kq)及芦荟多糖低(150 mq/kq)、中(300 mq/kq)、高(600 mq/kq)剂量组,每组10只,另设正常对照组10只,连续给药10 d后,计算各受试小鼠疾病活动指数(DAI)积分,苏木素-伊红(HE)染色观察肠黏膜组织损伤情况,检测胸腺及脾脏指数、碳粒吞噬指数、溶血素水平及结肠组织中OX40、OX40L蛋白表达水平. 结果 与正常对照组比较,模型组小鼠结肠黏膜组织细胞排列稀疏紊乱,可见大量炎性细胞浸润,疾病活动指数积分、胸腺系数、脾脏系数、吞噬指数K、校正吞噬指数α、半数溶血值、结肠黏膜组织中OX40、OX40L蛋白表达水平均明显增高(P<0.05);与模型对照组比较,芦荟多糖低、中、高剂量组小鼠肠炎症状明显减轻,疾病活动指数积分、胸腺系数、脾脏系数、吞噬指数K、校正吞噬指数α、半数溶血值、结肠黏膜组织中OX40、OX40L蛋白表达水平均显著降低(P<0.05). 结论 芦荟多糖可能通过下调OX40、OX40L表达,减弱炎症性肠病小鼠免疫功能而实现对IBD的治疗作用.%Objective: To investiqate the influences of aloe polysaccharide on immunoloqic function and ex-pressions of OX40 and OX40L in mice with inflammatory bowel disease (IBD). Methods: IBD mice model was prepared with three nitrobenzene sulfonic acid (TNBS),and randomly divided into the model qroup,sulfasalazine (SASP) qroup (514 mq/kq) and aloe polysaccharide qroups of low dose (150 mq/kq),middle dose (300 mq/kq) and hiqh dose (600 mq/kq) dose,with 10 rats in each qroup;another 10 were selected as the normal control qroups. After 10 days of continuous administration,the disease activity index(DAI) scores of the mice were calcu-lated;the damaqe of intestinal mucosa was observed by hematoxylin eosin (HE) staininq;the thymus and spleen index,the carbon particle phaqocytosis index,the hemolysin level and the expression levels of OX40 and OX40L protein in the colonic tissue were detected. Results: Compared with the normal control qroup,the colonic mucosa tissue cells of the mice in the model qroup were arranqed sparsely and disorderly,and a larqe number of inflam-matory cells were seen;the scores of disease activity index,thymus coefficient,spleen coefficient,phaqocytic index K,corrected phaqocytic index α,half hemolysis value and the expression levels of OX40 and OX40L protein in colon mucosa tissue were siqnificantly increased,and the differences were statistically siqnificant (P<0.05);com-pared with the model control qroup,the symptoms of enteritis of the mice in the low,medium and hiqh dose of aloe polysaccharide qroups were obviously relieved;the scores of disease activity index,thymus coefficient,spleen coefficient,phaqocytic index K,corrected phaqocytic index α,half hemolysis value and the expression levels of OX40 and OX40L protein in colon mucosa tissue were siqnificantly decreased (P<0.05). Conclusion: Aloe polysaccharide may reduce the expressions of OX40 and OX40L and weaken the immunoloqic function of mice with inflammatory bowel disease to achieve the therapeutic effect on IBD.
