电针次髎穴对慢性膀胱过度活动症模型大鼠尿动力学及脊髓背角SP和CGRP影响

         

摘要

[Objective] To investigate the urodynamic effects of electroacupuncture at Ciliao (BL 32) on overactive blad-der( OAB) in rats induced by cyclophsphamide( CYP)and expressions of calcitonin - gene related peptide( CGRP)and SP in dorsal horn of spinal cord. [ Methods ] Twenty five Sprague - Dawley rats were randomly divided into: control group (n=5), chronic OAB group (n = 10), chronic OAB + electroacupuncture group (n = 10). OAB model was established by intraperitoneal injection (I. P. I) of cyclophsphamide, continuous cystometry was performed to detectinter - contraction interval, basal, threshold, micturition voiding pressure before and after electroacupuncture. The expressions of CGRP and SP in the dorsal horn of spinal cord were delected by ininuinohistochemical method. [Results] Compared with control group, inter - contraction interval was decreased (P <0.05) and basal pressure was increased (p<0.01) in both OAB and ele-ctroacupuncture group before acupuncture. While basal pressure was decreased and inter - contraction interval was increased significantly in acupuncture group compared with those in control group (P < 0. 01 ). Comparison with control group, the mean optical density (OD) value of SP and CGRP positive products in the dorsal hom of sacral cord in OAB group was increased significantly (P<0.05), while compared with OAB group, the mean OD value of SP and CGRP in acupuncture group was decreased (P < 0. 05). [Conclusions] Electroacupuncture at CiLiao could inhibit the overactive bladder induced by cyclophsphamide in rats by decreasing the SP and CGRP expressions in the dorsal horn of spinal cord.%[目的]探讨电针次髎穴对环磷酰胺( cyclophsphamide,CYP)致慢性膀胱过度活动症(overactive bladder,OAB)模型大鼠尿动力学及脊髓背角p物质(substance P,SP)和降钙素基因相关肽(calcitonin - gene related peptide,CGRP)的影响.[方法]雌性大鼠25只随机分为对照组5只,OAB模型组10只,OAB+电针(电针组)10只.其中模型组和电针组动物腹腔内注射环磷酰胺诱导OAB,应用膀胱测压技术动态观察电针次髎穴后排尿间期、基础膀胱压、膀胱最大充盈压及最大排尿压的变化;应用免疫组织化学方法定量观察电针后脊髓背角SP和CGRP表达变化.[结果]电针组和模型组的排尿间期与对照组相比明显缩短(P<0.05),基础膀胱压明显升高(P<0.01).电针后电针组的基础膀胱压较模型组明显降低(P<0.01),排尿间期较模型组明显延长(P<0.01).与对照组相比,模型组脊髓后角SP及CGRP表达明显升高(P<0.05);与模型组相比,电针组的脊髓后角SP及CGRP表达降低(P<0.05).[结论]电针次髎穴能抑制环磷酰胺诱导的大鼠膀胱过度活动,降低脊髓后角SP和CGRP的表达,电针次髎穴治疗OAB可能与抑制膀胱传入神经的兴奋性及传入递质的释放与合成有关.
