Objective To investigate the clinicopathological significance of WW domain-containing oxidoreductase(WWOX) mRNA expression in colorectal carcinoma and paracancerous tissues and discuss the potential correlation between WWOX and p53 gene on transcription level. Methods The specimens from 30 cases of colorectal carcinoma and paracancerous tissues were collected to detect the mRNA expression of the WWOX gene and the exons 7 and 8 of p53 gene by reverse transcription polymerise chain reaction(RT-PCR) analysis in colorectal carcinoma and paracancerous tissues. Results The mRNA expression of WWOX and p53 gene in the tumor tissues was lower than that in the adjacent tissues, and the difference was significant. The mRNA levels of WWOX and p53 gene in tumor tissues and adjacent tissues were positively related. The low expressions of WWOX and p53 mRNA in colorectal carcinoma tissues were also positively correlated. The clinicopathological analysis showed that the WWOX mRNA expression related with histological grading, invasion depth and lymph node metastasis of carcinoma statistically. Conclusion Alterations of the WWOX gene may be involved in colorectal carcinoma. WWOX gene may prove to be a useful marker for predicting the biological aggressiveness and a target for diagnosis and gene therapy in colorectal carcinoma.%目的 研究抑癌基因WWOX mRNA在大肠癌及癌旁组织中的表达及与临床病理学意义,并在转录水平探讨WWOX与p53的相关性.方法 用半定量RT-PCR方法检测30例大肠癌组织及相应癌旁组织WWOX mRNA与p53第七、八外显子mRNA表达情况,就WWOX mRNA表达水平与临床变量之间进行分析.结果 WWOX mRNA、p53 mRNA在大肠癌中的表达显著低于其癌旁组织,差异有统计学意义,WWOX mRNA与p53 mRNA在大肠癌及癌旁组织中的表达呈正相关,WWOX mRNA与p53mRNA在大肠癌组织中的低表达亦呈正相关.临床病理研究发现WWOX mRNA表达与大肠癌分化程度、肿瘤浸润深度、淋巴结转移有统计学意义.结论 WWOX mRNA表达改变可能参与大肠癌的发生,可作为预测大肠癌生物侵袭性的有效指标之一,成为大肠癌诊断和基因治疗的一个靶点.
