硫化氢对脂多糖致大鼠 ARDS 时肺组织 NF-κB 表达的影响

         

摘要

目的:探讨外源性硫化氢对脂多糖(LPS)致大鼠急性呼吸窘迫综合征(ARDS)的保护作用机制。方法将30只健康雄性 SD 大鼠随机分为3组:对照组(Control)、模型组(LPS)和硫化氢干预组(NaHS+ LPS),每组10只。静脉注射 LPS 建立 ARDS 模型,ELISA 法检测大鼠血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量的变化;免疫组化检测肺组织中核因子-κB(NF-κB)的表达变化。结果模型组较对照组大鼠血清中 TNF-α、IL-6含量增多,肺组织中 NF-κB 表达增加(均 P <0.05);硫化氢干预组与模型组相比,血清中 TNF-α、IL-6含量降低,肺组织中 NF-κB 表达减少(均 P <0.05)。结论外源性硫化氢对脂多糖致ARDS 大鼠具有一定的保护作用,其机制可能与降低 TNF-α、IL-6含量、减少 NF-κB 表达有关。%Objective To investigate the effect of hydrogen sulphide on the expression of NF-κB in rats with acute respiratory distress syndrome caused by lipopolysaccharide (LPS). Methods 30 SD male rats were randomly divided into 3 groups: the control group, the LPS group and the NaHS + LPS group. The content of TNF-α and IL-6 in serum and the expression of NF-κB in lung tissues were tested. Results Compared with the control group, the content of TNF-α and IL-6 in serum and the expression of NF-κB in lung tissues increased significantly in the LPS group (all P < 0. 05). Compared with the LPS group, the content of TNF-α and IL-6 in serum and the expression of NF-κB in lung tissues decreased obviously in the NaHS + LPS group (all P < 0. 05). Conclusion Hydrogen sul-phide has protective effect to rats with acute respiratory distress syndrome induced by LPS, which is probably correla-ted with the decrease of TNF-α and IL-6 and the low expression of NF-κB.
