煤焦油沥青烟气诱导 SD 大鼠肺癌动物模型的实验研究

         

摘要

Objective To establish and evaluate coal tar pitch(CTP)- induced SD rat model of lung cancer. Methods Fifty SD rats were put into the contamination box and breathed CTP smoke for 3 hours per day. The state of the rats were observed and recorded. The rats were sacrificed after 32 weeks. The lungs of rats were removed and pathological examination of the lung was made. The incidence of lung cancer was an-alyzed. Results In the early stage of CTP smoking,some rats had irritable cough;but food taking,activity and the weight gain were normal in most of the rats. In the later stage of the experiment,the rats had weight lose and listlessness,and 4 rats died at the end of the experiment. Differ-ent types of lung cancer were found in 36 rats(69. 57% ). Conclusion CTP smoking can be used to induce SD rat model of lung cancer. It had advantages of high success rate,low cost,simple and feasible.%目的:采用煤焦油沥青(CTP)烟气吸入法诱导 SD 大鼠肺癌疾病模型,评价该法造模的优缺点以及对其效果进行观察。方法将50只 SD 大鼠放入染毒柜内,每日给予相同浓度的 CTP 烟气熏蒸,每天持续3 h。每周观察并记录小鼠的生长、活动、饮食和健康状况,并称量体重。32周后处死全部存活大鼠,取肺组织检查肿瘤发生情况,并对标本常规石蜡制片,HE 染色,病理学方法检查大鼠肺癌发生情况。结果个别实验鼠在实验早期偶尔有刺激性咳嗽表现,大部分大鼠精神状态较好,饮食、活动以及体重增长基本正常。但在实验后期大鼠体重均有所下降,精神萎靡、消瘦。实验结束时存活大鼠46只,4只死亡。存活大鼠中36只罹患不同类型肺癌,肺癌发病率为69.57%。结论选用CTP 烟气吸入法可以诱导大鼠肺癌疾病模型,且此方法实验鼠肺癌发病率高,实验成本低,简单易行,是实验室获取肺癌动物模型的较理想选择。
