骨桥蛋白和基质金属蛋白酶-3、10在结肠癌组织中的表达及意义

摘要

Objective To study the expression of osteopontin,matrix metalloproteinase-3 and matrix metalloproteinase-10 in colonic carcinoma and explore the relationship between their expression and clinicopathologic features.Methods Expression of OPN,MMP-3 and MMP -10 were studied via immunohistochemistry in sprcimens resected from 60 patients with colonic carcinoma and 26 paraneoplastic normal mucosa (PNM).And to observe the difference between the three indicators and the olinioopathological characteristics of colon cancer.Results Differences in expressions of OPN,MMP-3 and MMP-10 were significant between cancer mucosa and PNM (P ＜ 0.01).Expressions of MMP-3 and MMP-10 were both correlated with tumor size,depth of tumor invasion,LN metastasis and TNM stage (P ＜0.05).Expressions of OPN were correlated with LN metastasis (P ＜ 0.05),and those of MMP-3 and MMP-10 were positively correlated with that of OPN (P ＜ 0.05).Conclusion The abnormal expressions of OPN,MMP-3 and MMP-10 are closely related with the invasion and metastasis,and that can be used as important parameters for the judgment of biological and prognosis of colonic carcinoma.%目的 研究骨桥蛋白(OPN)和基质金属蛋白酶-3(MMP-3)、基质金属蛋白酶-10(MMP-10)在结肠癌中的表达及其意义.方法 采用免疫组织化学方法检测OPN、MMP-3及MMP-10在60例结肠癌和26例癌周正常黏膜组织中的表达,并观察三者与结肠癌临床病理特征之间的关系.结果 结肠癌组织中OPN、MMP-3及MMP-10的表达率明显高于癌周正常黏膜组织,差异均有显著性(P ＜0.01);MMP-3及MMP-10的表达均与肿瘤的大小、浸润深度、淋巴结转移及TNM分期有关(P＜0.05);OPN表达与淋巴结转移有关(P ＜0.05);MMP-3、MMP-10的表达与OPN的表达呈正相关(P＜0.05).结论 OPN、MMP-3及MMP-10的异常表达与结肠癌的侵袭转移密切相关,可能作为临床评估结肠癌进展及肿瘤预后的指标.
