间充质干细胞通过调节趋化因子及血管内皮生长因子异常表达减轻大鼠碱烧伤角膜的机制研究

         

摘要

目的 分析趋化因子及血管内皮生长因子异常表达与大鼠碱烧伤角膜的关系及间充质干细胞的调节作用,为相关治疗提供参考.方法 50只大鼠(100眼)随机分为5组:对照组、碱烧伤角膜模型组、间充质干细胞小剂量组、间充质干细胞中剂量和间充质干细胞高剂量组(n=20).除对照组大鼠给予等量的生理盐水处理外,其余各组大鼠采用1mol/L的NaOH溶液复制大鼠碱烧伤角膜模型,间充质干细胞治疗组采用对应剂量(0.25×106,0.5×106,1.5×106)进行治疗,对照组大鼠给予等量的生理盐水.Western Blotting法分析各组大鼠角膜组织中趋化因子CC趋化因子配体(CCL)2、CCL3、CCL8、CCL14、趋化因子基质细胞衍生因子-1受体4(CXCR4)、基质细胞衍生因子1(SDF1)、VEGF、VEGFR及白细胞介素的表达变化.结果 碱烧伤大鼠角膜CCL2、CCL3、CCL8、CCL14、IL-1β、IL-2及IL-6、VEGF及VEGFR表达明显增强,而SDF1和CXCR4表达明显减弱,且与对照组相比差异有显著性(P<0.05);经间充质干细胞治疗后上述蛋白的异常表达得到部分恢复(P<0.05),且呈剂量依赖性.结论 间充质干细胞能够通过调控趋化因子及血管内皮生长因子表达发挥改善大鼠碱烧伤角膜的作用,且作用具有剂量依赖性效果.%Objective To explore the effect of mesenchymal stem cells on alkali burned corneal in rats and the involvement of chemotactic factor and VEGF. Methods Clean rats were divided into 5 groups,control,model and mesenchymal stem cells intervention group. The alkali burned corneal rats were duplicated by the NaOH solution at 1M concentration. The mesenchymal stem cells intervention group rats were treated with mesenchymal stem cells at 0.25×106,0.5×106,1.5×106 . The content of CCL2,CCL3,CCL8,CCL14,CXCR4,SDF1,VEGF and VEGFR were assayed by Western Blotting. Results The results showed that the expression of CCL2,CCL3,CCL8,CCL14,IL-1β,IL-2, IL-6,VEGF and VEGFR in intervention group were increased,while the SDF1 and CXCR4 were decreased significantly in model group (P<0.05). The results also revealed that these abnormalities were normalized greatly after the medication of mesenchymal stem cells (P<0.05). Conclusion Mesenchymal stem cells are effective in the alleviation of alkali burned corneal rats which maybe related with the medication of chem-otactic factor and VEGF and its effects show a dose dependent manner.
