Fatty acid synthase (FAS) is reported as a potential new therapeutic target for obesity.However,there are limited known FAS inhibitors presently.Our study showed that the extract of oiltea shell (EOS) inhibited FAS activity with strong reversible fast -binding inhibition.40% ethanol was the optimal solvent for the extraction,and the half inhibition concentration ( IC_(50) ) of EOS to FAS overall reaction was 2.02μ g/mL.We also found the 70%ethanol extract of oiltea seed residue (EOSR) inhibited FAS activity with irreversible slow-binding inhibition.In the presence of 0.31 mg/mL of EOSR,the FAS activity inhibition was led to over 85% in 2 min.These results showed that FAS inhibitor from oiltea seed had stronger inhibition ability than the known inhibitors.It is expected to be a new effective FAS inhibitor,and may be of great value for the study on FAS mechanism and the control of obesity.%脂肪酸合酶(FAS)被认为是治疗肥胖症的潜在靶点.研究表明:油荼壳提取物对FAS具有很强的快结合可逆抑制作用.萃取的最佳溶剂为40%乙醇水溶液,此提取物的半抑制质量浓度为2.02μg/mL.而油茶饼70%乙醇提取物则具有很强的慢结合不可逆抑制作用,在0.31 mg/mL下2 min内FAS失活超过85%.结果表明:油荼壳提取物与油荼饼提取物分别在快结合抑制与慢结合抑制上显示出很强的抑制能力,预计为新的抑制剂,对研究FAS的作用机理与防治肥胖症的应用上可能具有重要的价值.
