钾盐胁迫下苦马豆愈伤组织生理指标研究

             

摘要

苦马豆为重要的耐盐抗旱药用豆科植物.作者研究了苦马豆愈伤组织在相同浓度NaCI盐和不同钾盐胁迫的生理效应.以苦马豆子叶外植体在添加1.79mg/L2,4-D和1.15mg/LKt的MS培养基上诱导的愈伤组织为材料,经相同盐浓度NaCl(200mmol/L)不同钾盐(KCl、KNO,或K2SO4)相同浓度([K^+]=150mmol/L)条件下胁迫,测定其在不同时间(0,3,6,9d)胁迫处理后愈伤组织中可溶性蛋白含量、SOD和POD活性及脯氨酸含量等生理指标.结果显示KN03胁迫组的积极响应度大于含KCl和K2SO4组,含K2SO4组最弱.表明浓度150mmol/LKNO、更利于盐胁迫抗性愈伤组织筛选.%Swainsona salsula Taubert is an impotant legume for its high tolerance of drought and high salts. In this study the physiological response of Swainsona salsula Taubert callus to salts stress was investigated by treatirig with the same concentration of NaC1 and different potassium salts. Callus was induced from cotyledons explants on MS medium supplemented with 1.79 mg/L 2,4 - D and 1.15 mg/L Kt for 36 days, and then were subcultured onto the same medium by adding 200 mmol · L^-1 NaCl with different potassium salt ( [ K^+ ] = 150mmol·L^-1 ) for salt stress. The physiological characters, including the activities of SOD and POD, contents of soluble protein and pra- line, were tested after 0, 3, 6, 9 days. The results showed that the salts stress response of callus from the medium with KNO3 (150mmol · L^-1) was more positive than the callus on medium with K2SO4 or KCl, and the callus on medium with K2SO4. was weakest.
