降低ATM的表达促进儿童AML细胞增殖和G0/G1向S/G2期细胞转化

         

摘要

目的 研究ATM基因在急性髓系白血病细胞(AML)中的表达对细胞增殖和细胞周期的影响.方法 采用定量PCR检测儿童AML白血病细胞中ATM基因的表达情况.采用siRNA转染AML白血病细胞株NB4及K562降低ATM基因的表达.采用CCK-8计数法检测ATM基因对人AML细胞系增殖的影响,采用流式细胞术分析细胞周期情况.结果 ATM基因在儿童AML白血病细胞中表达明显降低,与对照组相比差异有显著性(P<0.05).CCK-8检测结果显示,降低ATM基因的表达可以促进白血病细胞增殖,且可以促进白血病细胞细胞周期转变,促进GO/G1期细胞向S及G2期细胞转变.结论 儿童AML白血病细胞中ATM基因低表达,抑制ATM基因表达可以促进AML细胞增殖及G0/G1期细胞向S及G2期细胞转变.因此,ATM基因在AML中起“抑癌基因”作用.%Objective To investigate the expression of ATM in children with acute myeloid leukemia (AML) cells and explore its impact on proliferation and cell cycle.Methods The expression of ATM in children AML ceils was detected by quantitative polymerase chain reaction (qPCR).Moreover,cell cycle and cell proliferation were evaluated in cell lines (NB4 and K562) by using CCK-8 and flow cytometry after transfected with ATM siRNA.Results We found that ATM was significantly decreased in children AML cells,compared with that in control group (P < 0.05).Through reducing expression of ATM in cell lines by siRNA transfection,cell proliferation was significantly enhanced and the ratio of S-phase and G2-phase cells was increased.Conclusions Downregulation of A TM in children AML cells can promote AML cell proliferation and transition from G1-phase to S-phase.Therefore,ATM may act as a tumor suppressing gene.
