Objective To explore the effect of scytonemin on proliferation of prostate cancer cell line 22RV1 and activity of PLK1. Methods At different concentrations (1,2,3 and 4 jjunol/L) of scytonemin,MTT was used to exam the cell proliferation,scintillation counter was used to observe the activity of PLK1. Results The cell proliferation and the activity of PLK1 decreased significantly with the increasing concentration of scytonemin. The PLK1 activity in 3 and 4 junol/L scytonemin groups was significantly lower than the control group( P < 0.01). Conclusion The proliferation of prostate cancer cell 22RV1 and the activity of PLK1 could be inhibited by certain concentration of scytonemin.%目的 探讨scytonemin对前列腺癌细胞系22RV1的增殖及PLK1蛋白活性的影响.方法 使用MTT法检测不同浓度scytonemin(终浓度分别为1、2、3、4μmol/L)对前列腺癌细胞系22RV1形态及增殖活性的影响,应用激酶活性测定法检测不同浓度scytonemin对PLK1活性的影响.结果 随着scytonemin作用浓度的增加,细胞增殖能力逐渐下降,同时PLK1蛋白活性下降,与对照组相比,scytonemin浓度为3μmol/L和4mol/L时,差异具有统计学意义(P<0.01),其余各浓度组则差异均无统计学意义.结论 scytonemin达到一定浓度后,可以抑制前列腺癌细胞系22RV1的增殖活性及PLK1活性.
