草麻黄补体抑制成分在大鼠急性脊髓损伤中的作用

         

摘要

Objective To investigate the effect of complement inhibiting component of Ephedra sinica on acute spinal cord injury in rats.Methods The inhibiting component of complement in Ephedra sinica was isolated and purified by multiple precipitative steps and thin layer chromatography,the activity was evaluated.Induction of spinal cord injttry was performed following a modified Allen's weight-drop method.CH50 method was used for Measurement of complement hemolytic activity.ICAM-1 mRNA level was evaluated by real time PCR technique.Results CH50 and ICAM-1 mRNA levels were significantly reduced in the Ephedra sinica group compared with the Control group at 12h,1,3,7,14d after spinal cord injury.On the 7th and 14th day,the levels of latency and amplitude in Ephedra sinica group were significantly better than those of Control group (P < 0.01 ).Conclusion The inhibiting component of complement in Ephedra sinica significantly reduced immunological inflammation after spinal cord injury, and played an important protective function in secondary spinal cord injury.%目的 探讨草麻黄补体抑制成分在大鼠急性脊髓损伤中的作用.方法 应用多步沉淀和薄层层析方法进行草麻黄补体抑制成分的提纯并进行活性鉴定;参照改良的Allen's重物打击法制备大鼠脊髓损伤模型;应用CH50法测定血清总补体溶血活性;应用实时PCR技术检测细胞间黏附分子1(ICAM-1)mRNA的表达;采用运动诱发电检测评定脊髓损伤后的神经功能.结果 在伤后12 h、1 d、3 d、7 d、14 d各个时间点,实验组CH50比值和ICAM-1 mRNA表达均低于对照组(P<0.01).伤后7 d和14 d,实验组脊髓运动诱发电位潜伏期和波幅显著优于对照组(P<0.01).结论 草麻黄补体抑制成分能够显著减轻脊髓损伤后的免疫炎性反应,在继发性脊髓损伤中起到重要的保护作用.
