紫柏凝胶抑制宫颈癌Siha细胞增殖和诱导凋亡的实验研究

摘要

Objective To study the Zibai gel on proliferation inhibition and apoptosis of HPV positive Siha cells. Methods Siha cells in logarithmic growth phase were selected, divided into different concentrations groups (15, 7.5, 3.25 mg/mL) of Siha cells, and Siha cells without medicine was used as control group. Cell growth inhibition was detected by MTS. Trypan blue staining was used to detect cell survival rate. Acridine orange / ethidium bromide (HOECHST 33342) staining method was used to detect the changes of apoptosis. Results MTS assay: The inhibitory effect of each group "Zibai gel" on Siha cell growth were different, the 3.125 mg/L dose group was statistically significant compared with the control group (P < 0.05), IC50 (15±2.1) mg/mL; trypan blue staining: Zibai gel has a certain inhibitory effect on the growth of Siha cells, the inhibition rate of cell growth in each concentration group was higher than that in control group (P < 0.05); action of Zibai gel on Siha cells leads to a qualitative change between cells, loosening of cytoplasm, increase of cytoplasmic granules, condensation of chromatin and fragmentation of cytoplasm; HOECHST 33342/PI staining: the apoptosis of the cells was observed by fluorescence microscope. Conclusion Zibai gel can inhibit cervical cancer Siha cell proliferation and induce apoptosis.%目的 研究紫柏凝胶对HPV阳性宫颈癌细胞Siha的增殖抑制和凋亡诱导作用.方法 选取对数生长期Siha细胞,采用不同浓度(15、7.5、3.25 mg/mL)组,不加药物Siha细胞为对照组.MTS检测细胞生长抑制,台盼蓝染色检测细胞存活率,HOECHST 33342/PI染色法检测凋亡细胞形态学改变.结果 MTS法检测,各组紫柏凝胶对Siha细胞的生长均有不同程度的抑制作用,3.125 mg/mL剂量组与空白组相比有统计学意义(P<0.05),IC50为(15±2.1)mg/mL;台盼蓝染色,紫柏凝胶对Siha细胞生长有一定抑制作用,各浓度组细胞生长抑制率均高于对照组(P<0.05);紫柏凝胶作用Siha细胞后细胞间质变松,细胞质颗粒增多,染色质凝聚,分块,胞质浓缩;HOECHST 33342/PI染色,荧光显微镜下观察到紫柏凝胶引起细胞发生凋亡.结论 紫柏凝胶能抑制宫颈癌Siha细胞的增殖和诱导其凋亡.
