艾灸对FCA诱导的AA模型大鼠心肌超微结构及血清代谢组学的影响

         

摘要

Objective To study the effects of moxibustion on myocardial ultrastructure and serum metabolism in AA model rats induced by FCA.Methods 80 rats were randomly divided into NC group, TPT group, MC group and moxibustion group, 20 rats in each group. Respectively inject rat's right rear toes with intradermal FCA which induced inflammation (except NC group), and give inflammation treatment after 3 days, NC group and MC group were not included. TPT group was given TPT suspension once a day according to the weight, while moxibustion group was given moxibustion therapy, once a day. After 15 days' treatment, the metabolism technology was used to detecte the changes of endogenous metabolites in serum of rats. And observed the changes of rats myocardial ultrastructure by electron microscopy.Results Moxibustion and TPT can obviously improve AA model rats paw swelling and AI, which the improvement of the AI in moxibustion group was more obvious (P<0.05); Compared with MC group, the moxibustion and TPT can protect rats myocardial function, the injury more small in the moxibustion group; Based on PCA, finding important ucrose metabolic marker is glucose, alanine, ornithine, BHT, arachidonic acid,and the metabolic pathway of information disturbance mainly involved: carbohydrate metabolism, amino acid metabolism, intestinal flora and fatty acid metabolism.Conclusion Moxibustion can improve inflammation and protect myocardium on AA rat, and the regulation of amino acids and intestinal flora and fatty acid metabolism may be the specificity of the moxibustion treatment of RA.%目的 艾灸对弗氏完全佐剂(CFA)诱导的佐剂性关节炎(AA)模型大鼠心肌超微结构及血清代谢组学的影响.方法 将80只大鼠按随机数字表随机分成正常对照(NC)组、雷公藤多苷片(TPT)组、模型对照(MC)组和艾灸组,每组20只,分别于大鼠右后足跖皮内注射FCA致炎(NC组除外),致炎3 d后NC组和MC组予以生理盐水,TPT组按体质量给予TPT混悬液,艾灸组予以艾灸治疗,1次/d,连续治疗15 d后,采用代谢组学方法,观察大鼠血清中内源性代谢物的变化.用电镜观察大鼠心肌线粒体超微结构的变化.结果 艾灸与TPT均能明显改善AA模型大鼠的足跖肿胀度及关节炎指数(AI),其中艾灸组大鼠AI改善更为明显(P<0.05).从心肌超微结构图来看与模型组相比,艾灸及TPT均有保护AA模型大鼠心肌作用,而艾灸组心肌的结构损伤更小.从血液代谢组学研究来看,依据PCA结论,发现变化过程的代谢标志物为葡萄糖、丙氨酸、鸟氨酸、对甲酚、花生四烯酸、蔗糖,其中扰动的代谢通路主要涉及糖类代谢、氨基酸代谢、肠内菌群及脂肪酸代谢.结论 艾灸能改善AA的炎症反应同时能够保护心肌,调控氨基酸、肠内菌群及脂肪酸代谢可能是艾灸治疗RA的特异性体现.
