siRNA 干扰 TRIM29基因对人肺癌 NCI-H520细胞株增生及迁移能力的影响

             

摘要

目的:探讨 TRIM29基因沉默对肺癌细胞株 NCI-H520增生和迁移能力的影响。方法将针对 TRIM29的 siRNA 导入NCI-H520细胞,用 real time PCR 和 Western blotting 法分析 TRIM29基因及蛋白表达情况,MTT 法和 Transwell 小室法检测其增生、迁移能力。结果 NCI-H520细胞转染48 h 后,与空白组及对照组相比,TRIM29 siRNA 转染组 TRIM29 mRNA 和蛋白表达均明显下调,细胞生长明显减慢,细胞迁移能力下降。结论 siRNA 干扰下调 TRIM29基因表达能抑制肺癌细胞 NCI-H520的增生和迁移能力。%Objective To study the effects of TRIM29 gene on the proliferation and migration of human lung cancer cell line NCI-H520. Methods We transfected TRIM29 siRNA into NCI-H520 cells. Real time reverse transcriptase polymerase chain reaction and Western blotting assay were employed to determine TRIM29 messenger ( m) RNA and protein expressions. MTT assay was used to investigate the proliferation of the cells, and the cell migration was evaluated using a transwell migration assay. Results The mRNA and protein levels of TRIM29 were significantly downregulated at 48 h after transfection in TRIM29 siRNA group compared with the control groups. The cell proliferation and migration were significantly reduced. Conclusion siRNA targeting TRIM29 gene can inhibit the proliferation and migration of human lung cancer cell line NCI-H520.
