CDK5在血管性认知障碍大鼠海马中的表达

[ Abstract] Objective To investigate the change of expression and possible effect of cyclin-dependent protein kinase 5(CDK5) in hippocampal tissue of mice with vascular cognitive impairment(VCI).Methods Eighty male SD rats were randomly divided into three groups: middle cerebral artery occlusion ( MCAO) group, sham-operated group and normal group.MCAO Rats model were made by intraluminal method.The behavioral tests were done at 30 days after model builded,and VCI rats were selected.Then the CDK5 mRNA and CDK5 protein expression were examined by RT-PCR and Western-blot.Results The expression of CDK5 mRNA and CDK5 protein in VCI group was significantly higher than sham-operated group and normal group ( P <0.05).Conclusion CDK5 contributes to VCI in rats.%目的:探讨细胞周期素依赖性蛋白激酶5(CDK5)在血管性认知障碍(VCI)大鼠海马组织中的表达变化及其可能作用。方法将雄性 SD 大鼠80只随机分为大脑中动脉梗塞(MCAO)组、假手术组及正常对照组,采用改良线栓法建立 MCAO 大鼠模型,30d 后进行水迷宫实验评价并筛选 VCI 大鼠,分别应用RT-PCR、Western-blot 方法检测各组大鼠海马组织 CDK5 mRNA 及 CDK5蛋白的相对表达量。结果VCI 大鼠海马组织 CDK5的表达水平明显升高(P<0.05)。结论CDK5可能参与了大鼠 VCI 的发生。
