丁苯酞对大鼠局灶性脑缺血再灌注后NF-κB与p38MAPK表达的影响

     

摘要

Objective To observe the effect of dl-3n-butylphthalide (NBP) on the expression of nuclear factor-kB (NF-κB) and p38 mitogen activated protein kinase (p38MAPK) in the rat of focal cerebral ischemia-reperfusion (I/R). Method One hundred male SD rats were randomly divided into sham operation group, I/R group, NBP low-dose (20mg·kg-1) group, NBP middle-dose (40mg·kg-1) group and NBP high-dose (80mg·kg-1) group. The rat model of focal brain I/R was established with the suture-occluded method introduced by Zea Longa. The cerebral infarct volume, histological changes of brain and the expression of NF-κB and p38MAPK were detected. Results There were significant cerebral infarct volume, histological changes and elevated expression of p-NF-κB and p-p38MAPK in I/R group compared with sham operation group. The the expression of p-NF-κB and p-p38MAPK were significantly decreased (P<0.05) in NBP group compared with I/R group. Conclusion NBP can relieve the brain damage induced by focal I/R in rats, which may be correlated with the inhibition of expression of p-NF-κB and p-p38MAPK.%目的 探讨丁苯酞(NBP)注射液对大鼠局灶性脑缺血再灌注(I/R)损伤后核转录因子-κB (NF-κB)与p38丝裂原活化蛋白激酶(p38MAPK)表达的影响.方法 清洁级健康SD大鼠100只,根据随机分组法分成5组:假手术组(Sham组)、I/R组、NBP小剂量组(20mg·kg-1)、NBP中剂量组(40mg·kg-1)和NBP大剂量组(80mg·kg-1) 各20只.采用Zea-longa 线栓法制作大鼠局灶性IR 模型,使用TTC染色法检测各组大鼠的脑梗死体积,苏木精-伊红染色法检测各组大鼠的脑梗死病理变化,利用Western blotting方法检测各组大鼠的NF-κB与p38MAPK表达以及NBP 组大鼠经不同剂量NBP处理后的NF-κB与p38MAPK表达.结果 NBP 组和IR 组脑梗死体积明显高于Sham 组,NBP 组明显低于I/R 组(P<0.05) .I/R组和Sham组相比,细胞排列紊乱,细胞数目减少,细胞间隙增宽,甚至严重的出现核固缩.I/R组和NBP组比Sham组中的p-NF-κB与p-p38MAPK表达升高(P<0.05).结论 NBP能够抑制大鼠脑组织中磷酸化NF-B和磷酸化p38MAPK蛋白水平增加,减轻脑I/R损伤.
