眼针对大鼠急性脑缺血再灌注损伤脑组织ICAM-1表达的影响

     

摘要

Objective To observe the influence of eye acupuncture on the expression of brain intercellular cell adhesion molecules-1 (ICAM-1) in rats with cerebral ischemia and reperfusion injury for 24 hours, and discuss the effect mechanism of eye acupuncture to cerebral ischemia and reperfusion injury. Methods SD rats were randomly divided into blank group, sham-operation group, model group and eye acupuncture group. The rat model of cerebral ischemia and reperfusion was established by using suture method. The brain patholegical changes were observed with HE staining in rats of all groups. The expression of brain ICAM-1 was detected by using the methods of ELISA, immunohistochemistry, real-time fluorescence quantitative polymerase chain reaction ( RT-PCR) and Western-blot method. Results Compared with blank group, the content and expression of brain ICAM-1 increased significantly in model group. Compared with model group, the content and expression of brain ICAM-1 decreased significantly in eye acupuncture group. Conclusion One of effective mechanism of eye acupuncture for treating cerebral ischemia and reperfusion injury may be related to down-regulation of ICAM-1 expression.%目的 观察眼针对脑缺血再灌注模型大鼠再灌注损伤24 h后脑组织细胞黏附分子-1( ICAM-1)表达的影响,探讨眼针对脑缺血再灌注损伤的疗效机制.方法 SPF级SD大鼠随机分为空白对照组、假手术组、模型组、眼针组.采用线栓法制备大鼠脑缺血再灌注模型,HE染色观察各组大鼠脑组织病理变化,酶联免疫吸附法(ELISA法)、免疫组化、实时定量PCR、Western blot等方法检测各组大鼠脑组织中ICAM-1的表达.结果 与空白对照组比较,模型组ICAM-1的含量及表达明显增强;与模型组比较,眼针组ICAM-1含量与表达明显减弱.结论 眼针对脑缺血再灌注损伤疗效机制之一可能是下调机体ICAM-1的表达.
