艾烟冷凝物对大鼠肺泡巨噬细胞NR8383活性和吞噬功能的影响

     

摘要

Objective To observe the influences of moxa-smoke condensate in different doses on activity and phagocytosis of alveolar macrophages (NR8383) in rats.Methods In proliferation experiment,the doses of moxa-smoke condensate were designed as 0 g/L,0.004 g/L,0.008 g/L,0.012 g/L,0.016 g/L and 0.020 g/L.NR8383 was exposed to moxa-smoke condensate in the above-mentioned doses for 12 hours,24 hours,36 hours and 48 hours respectively.The proliferation of NR8383 was detected by applying CCK-8 method.In phagocytosis experiment,the doses of moxa-smoke condensate were designed as the same as the cell proliferation experiment.The phagocytic rate was detected by using flow cytometry 24 hours after NR8383 exposure.Results After NR8383 exposure for 12 hours,its survival rate was higher in all experimental groups than that in blank group but the difference was not statistically significant (P > 0.05).After 24 hours and 36 hours,the survival rate was higher in all experimental groups than that in blank group and the difference was statistically significant (P < 0.05) when the dose of moxa-smoke condensate was,respectively,0.004 g/L,0.008 g/L,0.012 g/L and 0.016 g/L,and the survival rate was lower in all experimental groups than that in blank group but the difference was not statistically significant (P > 0.05) when the dose of moxa-smoke condensate was 0.020 g/L.After 48 hours,the difference was statistically significant (P < 0.05) between blank group and all experimental groups except of 0.008 g/L group.After NR8383 exposed for 12 hours,24 hours,36 hours and 48 hours respectively,its survival rate showed a tendency of firstly increasing and then decreasing,and increased at first and decreased then as time extension and dose increase of moxa-smoke condensate.After NR8383 exposed for 24 hours,its phagocytosis was improved as dose increase of moxa-smoke condensate,and the difference was statistically significant (P < 0.005,corrected chi-square value) between blank group and all experimental groups except of 0.004 g/L group.Conclusion In a certain dose range,moxasmoke condensate can improve the activity and phagocytosis of NR8383 in rats showing a dosetime dependency.%目的 观察不同浓度的艾烟冷凝物对大鼠肺泡巨噬细胞NR8383活性和吞噬功能的影响.方法 在细胞增殖实验中,将艾烟冷凝物染毒浓度分别设计为:0、0.004、0.008、0.012、0.016、0.020 g/L,以上述不同浓度的艾烟冷凝物对NR8383进行细胞染毒,染毒时间分别为12、24、36、48h,然后采用CCK-8法对细胞增殖情况进行检测;吞噬功能实验中,艾烟冷凝物染毒浓度设计同上,对NR8383进行细胞染毒24 h后,采用流式细胞仪检测吞噬率.结果 艾烟冷凝物对大鼠肺泡巨噬细胞NR8383细胞进行染毒12 h后,各实验组的细胞存活率虽都高于空白对照组,但各实验组与空白对照组差异无统计学意义(P>0.05).染毒24、36 h后,实验组浓度为0.004、0.008、0.012、0.016 g/L时,各组细胞存活率不仅高于空白对照组,且各实验组与空白对照组差异有统计学意义(P<0.05),实验组浓度为0.020g/L时,其存活率虽低于空白对照组,但与空白对照组差异无统计学意义(P>0.05).染毒48 h后,除浓度为0.008g/L的实验组外,其余各组细胞存活率与空白对照组差异有统计学意义(P<0.05).艾烟冷凝物对NR8383细胞分别进行染毒12、24、36、48 h后,NR8383细胞存活率随着艾烟冷凝物浓度的升高均呈现先增加后降低的趋势,且随着作用时间的延长,细胞存活率亦随着艾烟冷凝物浓度的增高而先增加后降低.艾烟冷凝物对NR8383细胞分别进行染毒24 h后,NR8383细胞的吞噬功能随着艾烟冷凝物浓度的升高而增强.除浓度为0.004g/L的实验组外,其他各组与空白对照组差异均有统计学意义(P<0.005,校正卡方值).结论 在一定浓度范围内,艾烟冷凝物可提高大鼠肺泡巨噬细胞NR8383的活性和吞噬功能,并呈现一定的浓度及时间依赖性.
