高聚物反相填料在分离纯化莱菔硫烷中的应用

     

摘要

Sulforaphane has been separated and purified by polystyrene-based reversed phase chromatography (PST, 30 μm, 300mm × 10 mm ID). The preparative separation conditions were optimized and the optimum parameters were found to be 30% acetonitrile in water as the mobile phase, and a flow-rate of 2 mL/min in the iso-cratic elution mode. The highest purity of sulforaphane was 90% with a loading of 20 mg. The yield was more than 61%. With the use of overloading conditions and the heart-cutting method, the purity of sulforaphane exceeded 90%.%采用高聚物反相填料装填的半制备柱(PST,30 μm,300mm×10mm ID)对莱菔硫烷进行分离纯化,优化了色谱分离参数,确定最佳流动相为30%乙腈-水溶液,流速为2 mL/min等度洗脱,检测波长254 nm,进样量不超过20 mg时,收率在61%以上,纯度可达90%.采用中心切割的方式收集,获得的产品纯度可达90%以上.
