玉米Zma006292基因克隆与序列分析

         

摘要

[Objective] Maize is one of the most important crops in the world.The yield was reduced seriously by biotic and abiotic stresses.In plant NAC transcription factors play essential roles in the intricate mechanisms of signal transduction.They regulate many genes’ expression in different stress responses.In this study,an ATAF subfamily NAC transcription factor gene Zma006292 was isolated from the maize inbred line B73,and bioinformatics analysis was carried out.[Methods]The full-length eDNA of Zma006292 was amplified by RT-PCR.The sequence characteristics and evolutionary relationship were performed with multiple sequence alignment and phylogenetic analysis.The amino acids composition,conserved domain,sub-cellular localization and protein tertiary structure were analyzed with various bioinformatics softwares.[Result] The full-length cDNA of Zma006292 was 609 bp.%[目的]玉米是世界主要农作物之一,生物及非生物胁迫对玉米产量影响严重.植物复杂的逆境胁迫信号转导途径中,NAC转录因子通过调控诸多基因的表达发挥其抗逆功能.本研究从玉米自交系B73中分离得到ATAF亚家族NAC基因Zma006292,并对其进行序列分析.[方法]RT-PCR技术克隆基因全长cDNA,通过序列比对和构建系统进化树分析Zma006292氨基酸序列特征和进化关系.通过生物信息学预测氨基酸组成特征、保守结构域、亚细胞定位和蛋白三级结构.[结果]克隆得到Zma006292全长cDNA 609 bp.
