Twist在低氧微环境中宫颈癌上皮间质转化中的作用

         

摘要

Objective To investigate the role of twist on cervical cancer epithelial mesenchymal transition (EMT) in hypoxia microenvironment.Methods Hypoxia microenvironment was mimicked by using cobalt chloride.The morphologies and the migration ability of Hela cells in hypoxia microenvironment were observed by invert microscopy and scratch test.The expressions of hypoxia inducible factor-1α (HIF-1α),Twist and Vimentin proteins in Hela cells were determined by immunofluorescence.Results The mesenchymal morphological characteristics of Hela cells showed in hypoxia microenvironment.The migration percentage in the hypoxia group was (20.4±5.9)%,and was (5.2±1.7)% in the normoxia group (P<0.05).The expressions of HIF-lα,Twist and Vimentin of Hela cells in the hypoxia group significantly higher than those in the normoxia group (P<0.05).There were significantly positive correlations among them in the hypoxia group (P<0.05).Conclusion The expression of Twist increased in hypoxia microenvironment,and is highly correlation with EMT,Twist may induce EMT of cervical cancer through Twist pathway.%目的 研究Twist在低氧微环境中宫颈癌上皮间质转化(EMT)中的作用.方法 以二氯化钴处理Hela细胞模拟低氧微环境,细胞的形态变化、迁移能力分别应用倒置显微镜及划痕实验进行评价.Twist、Vimentin和低氧诱导因子-1α(HIF-1α)在Hela细胞中的表达应用细胞免疫荧光法进行检测.结果 低氧处理后Hela细胞获得了间质细胞的形态特征;Hela细胞低氧组迁移率为(20.4±5.9)%,常氧组为(5.2±1.7)%(P<0.05);低氧组Hela细胞HIF-1α、Twist、Vimentin表达均较常氧组显著增高(P均<0.05);低氧组Hela细胞中HIF-1α、Twist、Vimentin的表达两两均呈正相关(P<0.05).结论 低氧条件下Hela细胞中Twist高表达,这可能与宫颈癌EMT发生有关.
