[Objective] The research aimed to optimize the fermentation conditions of the engineering bacteria that secreted L-asparaginase into media. [Method] The ingredients of media were studied by orthogonal design. The effects of inducing conductions and aeration flow on enzyme production were studied by single-factor design. [Result] Fermentation media was composed of 8.0% corn steep liquor and 1.0% glycerol. 0.5 g/L lactose was added into media after culture 10 h to induce protein expression for 12 h. An appropriate aeration flow was 0.83 VVM. [Conclusion] This research provided basis for industrial production of extracellular expression L-asparaginase.%[目的] 优化胞外表达L-天冬酰胺酶工程菌的发酵条件.[方法] 用正交试验研究培养基的组成,用单因素试验研究诱导条件以及通气量对酶产量的影响.[结果] 发酵培养基采用8.0%玉米浆和1.0%甘油组成;诱导剂采用终浓度为0.5 g/L的乳糖,扩大培养后10 h加入,诱导时间为12 h,通气量为0.83 VVM.[结论] 该研究为胞外表达L-天冬酰胺酶的工业化生产提供了参考.
