A beagle dog was treated I.V. with 1,000 I.U./kg/day x 5. This dose regimen of L-Asparaginase gave plasma levels varying from 6.35 to 10.6 IU/ml of L-Asparaginase and the enzyme was still present in the plasma five days after the last dose. Interestingly no L-Asparaginase was present in the urine at any time during the dosage period. Plasma L-Asparagine was decreased and remained below the original level for 12 days after the last dose - - which was the last day on which samples were taken. In this beagle dog, as in previously studied beagles, L-Asparaginase did not produce a fatty liver nor was there any evidence of hepatotoxicity as indicated by BSP clearance, SGOT, SGPT, alkaline phosphatase, prothrombin time, and bilirubin. The drug regimen did not produce any significant alterations in hematology except for a transient decrease in WBC nor in BUN, electrolytes, serum proteins and cholesterol. (Author)
