菲律宾蛤仔提取物提取条件优化及其体外清除自由基的作用

         

摘要

[目的]研究菲律宾蛤仔全脏器提取物体外对自由基的清除作用,为其综合利用提供基础.[方法]分别采用邻二氮菲-Fe 氧化法、DPPH 反应法和邻苯三酚自氧化法,检测菲律宾蛤仔提取物体外对羟自由基(HO·)、DPPH·和超氧阴离子自由基(0·)的清除作用.[结果]以0.1 mol/L NaCl 溶液为浸提液,料液比为1:8,75 ℃水浴浸提100 min,提取液经透析浓缩,再以70%乙醇终浓度沉淀,提取物(PPS-O)的得率达到3.30%,总糖与可溶性蛋白含量分别为42.40%和14.40%.PPS-O 对HO·和 DPPH·清除作用的IC分别为3.100、3.500 mg/ml,对O·的清除率可达到39.4%.[结论]菲律宾蛤仔盐提取物具有一定的体外清除自由基作用.%[ Objective ] The technical basis of comprehensive utilization of the extracts from the all organs Ruditapes philippinarum for the free radical-scavenging in vitro was provided. [ Method ] The effect of the extracts from the Ruditapes philippinarum on the scavenging of on hydroxyl the radical( HO . ),the DPPH. radical and superoxide anion free radicals( O2 - . ) was measured with the method of phenanthroline respectively-Fe2+ oxidation, DPPH reaction and pyrogallol autoxidation. [ Result ] The rate of the extract(RPS-O) was 3.30%, the content of total sugar and soluble protein was 42.40% and 14.40% while the NaCl of 0. 1 mol/L was as extraction solution, the ratio solid to liquid was 1: 8,75 ℃ and the time of water-extracting was 100 minutes and then the extract solution was concentrated by dialysis concentrated and precipitated with the ethanol with the concentration of 70%. The IC50 ( the scavenging efficiency of RPS-0 on HO . and DPPH. was 3.100 and 3.500 mg/mi and the removal rate to O2 - . reached 39.4%. [ Conclusion]The salt extract from Manila clam had some role in free radical-scavenging.
