地衣芽孢杆菌BL03脂肪酶基因LIP的克隆及序列分析

摘要

[ Objective] To clone Bacillus licheniformis BL03 gene UP, and analyze the sequence. [ Method] The UP gene encoding lipase from B. licheniformis BID3 was amplified by PCR and cloned to the vector pMD19-T. [ Result] Nucleotide sequence analysis of the LIP gene revealed that the coding region was formed of one complete ORF including 615 base pairs. The cloned gene was 99. 67% similar with B. licheniformis LIP gene(GeneID:3098655) and the putative amino acid sequence was 99.02% similar respectively. [Conclusion] The cloned gene laid a foundation for the heteroiogous expression and relevant researches.%[目的]克隆地衣芽孢杆菌BL03的脂肪酶基因LIP并对其序列进行分析.[方法]以地衣芽孢杆菌BL03基因组核酸为模板,PCR扩增得到地衣芽孢杆菌脂肪酶基因,用pMD19-T载体克隆后进行PCR鉴定及序列测定和生物信息学分析.[结果]测序结果表明该片段完整开放阅读框大小为615 bp,与已发表的地衣芽孢杆菌LTP基因(GeneID:3098655)相比,核酸序列相似为99.67％;推导编码氨基酸序列相似性为99.02％.[结论]该基因的获得为其异源表达及相关研究奠定了基础.
