高效液相色谱-四通道紫外可见检测法测定不同产地谷精草中槲皮素含量

         

摘要

[ Objective] To establish a quantitative method of determination of quercetin in Eriocauion buergerianum ,and determine the content of quercetin in Eriocauion buergerianum from different growing areas. [ Method] High proficiency liquid chromatography ( HPLC) coupled with four-channel UV-visible detector was used, C18 chromatographic column, mobile phase was the buffer solution of methanol and phosphoric acid (0. 6% ) , the volume ratio of methanol and phosphoric acid (0.6% ) was 52= 48. Qualitative evaluation was done by retention lime and four-channel UV spectrum at the same time, and the detection wavelength were 254, 360, 365, 370 nm. Quantitative evaluation had done at the best sensitive wave of 254 nm. [ Result ] The linearity of quercelin was in the range of 5 - 30 μg/ml (R = 0.999 8), the average recovery was 99.56% , the RSD was 2. 30%. The content of quercetin in samples from diiferent growing areas was in the range of 29.5 - 92. 5 μg/ g, [ Conclusion ] The method is reliable, and can be used for quality control of Eriocauion buergerianum. The content of quercetin in samples from different growing areas has greater difference.%[目的]建立谷精草中槲皮素含量的测定方法,测定8个产地谷精草中槲皮素的含量.[方法]高效液相色谱-四通道紫外可见检测法,C18为色谱柱(250 mm ×4.6 mm,5 μm),流动相为无水甲醇-浓度0.6%磷酸(52∶48,V/V).通过保留时间结合四波长紫外光谱定性,定性检测波长为254、360、365和370 nm.选用灵敏度最高的波长254 nm定量.[结果]槲皮素在5~30μg/ml范围内线性关系良好(R =0.999 8),平均回收率99.56%,RSD为2.30%;不同产地谷精草中槲皮素含量范围为29.5~92.5μg/g.[结论]该方法测定结果可靠,稳定易行,重复性好,可用于谷精草中槲皮素的含量测定;不同产地谷精草中槲皮素含量差异较大.
