半夏中β-谷甾醇的含量测定

     

摘要

[Objective] The aim was to establish a HPLC method for determination of (I-sitosterol in Pinellia ternata( Thunb. ) Breit. [Method] The Shim-Pack VP-ODS C18 column(4.6 mm X 150 mm 5 |j.m)was used,and mobile phase was composed of CH30H. Detection wavelength was at 210 nm, the flow rate was 1. 0 ml/min, and column temperature was at 35 ℃. [Result] The linear response range from 0. 206 -2.060 μg/ml,r= 0.999 6. The recovery rate was 101.07% (re =5) ,RSD was 2.96%. [Conclusion] The HPLC method was accurate and suitable for the determination of β-sitosterol in Pinellia ternala( Thunb. ) Breit. .%[目的]建立高效液相色谱法测定半夏中β-谷甾醇的含量.[方法]色谱条件:色谱柱为岛津Shim-Pack VP-ODS C18(4.6 mm×150 mm,5μm),流动相为无水甲醇,流速为1.0 ml/min,检测波长为210 nm,柱温为35℃.[结果]β-谷甾醇在0.200~2.000 μg/ml浓度范围内线性关系良好,r =0.999 6,平均回收率为101.07% (n =5),RSD为2.96%;半夏样品中β-谷甾醇含量的平均含量为1.59mg/g.[结论]该方法分离度较好,可用于半夏药材中β-谷甾醇的含量测定.
