石榴乙烯反应因子PgERF1基因的克隆及序列分析

     

摘要

[目的]探讨乙烯反应因子在乙烯转导过程中的重要作用。[方法]克隆石榴ERF1基因,并对其进行序列分析。采用RACE和PCR技术,根据NCBI数据库中已知的ERF基因序列设计特异性引物,最终获得石榴ERF1的 cDNA序列。[结果] cDNA 序列全长1612 bp,可编码394个氨基酸。利用BLAST、DNAMAN及其他生物学信息分析软件,对该段序列进行结构分析,结果表明,其包含AP2结构域,并具有典型的ERF基因特点。经氨基酸同源性分析表明,石榴ERF1基因与其他物种ERF基因同源性较高;经系统进化树分析表明,其与欧洲栗、醉蝶花等物种亲缘关系较近。[结论]该研究成功克隆石榴ERF1基因,为后续的下游分析及园艺植物分子育种奠定基础。%Objective] The aim was to study the importance of ethylene-response factor in ethylene process.[ Method] The ERF1 gene was cloned from pomegranate fruit and sequence analysis was conducted.By PCR and RACE, according to the known ERF sequence in NCBI da-tabase, specific primers were designed to obtain the cDNA sequence of pomegranate fruit.[Result] The full-length DNA was 1 612 bp long, and encoded 394 amino acids.Blast, DNAMAN and other biological information analysis software were used to analyze the structure of se-quence.The results showed that the gene owns the typical characteristics of the ERF gene, which contains an AP2 domain.Homology analysis showed that, the ERF1 gene was hihgly homologous between pomegranate and other species, especilly the Castanes sativa and Tarenaya has-sleriana.[Conclusion] PgERF1 gene was successfully cloned, which will contribute to further analysis and molecular breeding of horticultural plants.
