籼型光敏核不育水稻Hs-1可育和不育幼穗mRNA的差异表达分析

         

摘要

The mRNAs from both fertile and sterile young panicle of photoperiod sensitive genic male sterile indica rice Hs-1 are analyzed by DD-PCR. The results are as following: Among 148 DD-PCR reactions, 10 reactions produce repeatable differentially expressed bands. In those 10 reactions, 4 of them each show 1~4 differential bands that are foundeither only in fertile young panicle or in both fertile and sterile young panicles. Each of the rest 6 reactions amplified more than 4 differential bands from both fertile and sterile young panicles. There are43 specific bands in total, among which 21 specific bands may be related to sterility. We suggest the key to isolate sterile genes is to anchor the start period of differential expression of these genes duringfertility transformation. It is considered that expression of sterilegenes may also result in transcription of other mRNAs and thus produce additional differential bands. It is important to further investigate the relationship between the differential bands and sterility.%本研究利用DD-PCR技术分析了光敏核不育籼稻Hs-1可育和不育幼穗mRNA的表达差异。结果表明:在148个DD-PCR反应中, 10个反应可扩增出有差异且可重复的cDNA片段,其中4个反应可扩增出1~4条差异片段,6个反应可扩增出4个以上的差异片段,且这些差异片段可只出现在可育幼穗中或同时出现在可育和不育幼穗中。共获得43个差异cDNA片段,其中21个可能与光敏核不育性相关。本研究认为,寻找光敏核不育基因在可育和不育条件下差异表达的起始发育时期是从mRNA水平分离这一基因的关键。光敏核不育基因的表达可能会引起其它一些mRNA的合成,从而产生较多的差异片段,进一步鉴定这些差异片段与不育的关系十分重要。
