运用16s rDNA PCR和实时荧光qPCR技术,对选育的低温沼气功能菌群中的产甲烷菌种群结构和数量进行了研究,并与常温沼气功能菌群进行了比较分析。在总共33个产甲烷菌分类单元中低温沼气功能菌群占有20个,低温沼气功能菌群的产甲烷菌多样性远高于常温沼气功能菌群。低温沼气功能菌群以对温度要求较低的中低温产甲烷菌为主,而常温沼气功能菌群则以中高温产甲烷菌占优。虽然两类菌群的产甲烷菌在多样性和种群结构方面存在着较大差异,但在产甲烷菌数量方面并没有明显的不同,这表明选育的低温沼气功能菌群和常温沼气功能菌群一样,已经形成了稳定的产甲烷菌种群。%Methanogens populations in low and normal temperature biogas functional microfloras were respectively investigated using 16s rDNA PCR and real-time qPCR techniques.The 16s rDNA clone library of methanogens showed a great difference in methanogens consortium diversity and structure between the two microfloras.A total of 33 OUTs of methanogens were identified,including 20 in low-temperature microflora and 13 in normal temperature microflora.Methanogens requiring low-medium temperature were found prevalent in low-temperature microflora and those active at moderate-high temperature occurred mainly in normal temperature microflora.However,the results of real-time qPCR showed that there was no significant difference in methanogen quantity between the two studied microfloras,indicating a stable methanogenic community had been built in both microfloras.
