不同剂量阿伦磷酸钠对去势大鼠股骨区域骨密度和力学性能的影响

             

摘要

Objective To explore the effect of different doses of alendronate on femoral region bone mineral density and me-chanical properties in an ovariectomized (OVX) rat model. Methods 40 female SD rats (6.5months) were randomly divided into 4 groups: control group,OVX group,alendronate A group (7mg/kg,once every two weeks),and alendronate B group (7mg/kg,per week),10 rats per group. Rats were recorded weight change every week. After treatment 12 weeks,the left femoral samples were collected to measure bone mineral density (BMD) and mechanical properties. Results There was no statistically significant among four groups on weight change in pre-/post-treatment. Compared with control group,the BMD of OVX group was obviously de-creased(P<0.05). There was no statistically significant among control,alendronate A and B groups. There was no statistically signif-icant between alendronate A and B groups on the BMD of proximal femur. Compared with alendronate A group ,the BMD of femoral shaft and distal femur in alendronate B group were obviously increased (P<0.05 for both). The cortical thickness,maximal load,yield load,and stiffness in femur collected from OVX group was significantly lower than that of control group (P<0.01 for all groups). Compared with OVX group,alendronate A and B groups significantly increase cortical thickness (P<0.05 for both),maxi-mal load(P<0.01 for both),yield load (P<0.05 for both) and stiffness (P<0.05 for both). No significant difference was indentified be-tween alendronate A and B groups. Conclusion The BMD and mechanical properties were abnormal in OVX rat. Alendronate A and B groups can increase femoral BMD and mechanical properties,especially,the BMD of femoral shaft and distal femur in alen-dronate B group was higher than that of A group.%目的:探讨不同剂量阿伦磷酸钠对去势骨质疏松大鼠区域性骨密度(BMD)和力学性能的影响。方法40只6.5个月龄SD雌性大鼠随机分成对照组、模型组、阿伦磷酸钠A组7mg/(kg·2周)、阿伦磷酸钠B组7mg/(kg·1周)。每周记录大鼠体重变化。治疗12周后,收集左侧股骨检测BMD和力学性能。结果各组大鼠治疗前后体重变化无统计学差异。模型组的总BMD低于对照组(P<0.01);对照组、阿伦磷酸钠A组与B组总BMD之间无统计学差异。股骨近端BMD,阿伦磷酸钠A组与B组相比无统计学差异。股骨干和股骨远端BMD,阿伦磷酸钠B组高于A组(均为P<0.05)。模型组的皮质骨厚度、最大载荷、屈服载荷、刚度均低于对照组(均为P<0.01)。阿伦磷酸钠A组和B组皮质骨厚度(均为P<0.05)、最大载荷(均为P<0.05)、屈服载荷(均为P<0.01)、刚度(均为P<0.05)高于模型组。阿伦磷酸钠A组和B组无统计学差异。结论去势大鼠均存在股骨不同区域BMD和力学性能下降,阿伦磷酸钠A组和B组均可提高股骨BMD和力学性能,阿伦磷酸钠B组对提高股骨干和股骨远端BMD优于A组。
