持久性大脑中动脉闭塞后远隔部位微管相关蛋白-2和胶质纤维酸性蛋白变化的实验研究

         

摘要

Objective To investigate the changes of MAP-2 and GFAP of the distal areas after long-time middle cerebral artery occlusion(MCAO).Methods 35 SD rats were divided into seven groups:the normal group;2 week group,and 1 month group after sham operation;2 week group,1 month group,3 month group and 6 month group after MCAO.Each group had five rats.Ischemic models were established by Tamura method to study the changes of thalamus,and substantia nigra reticulum(SNr).The neurons and gliocytes of the SNr were counted.Results It was observed that the areas of the thalamus and SNr had shrunk,and that the area,length and number of neurons in SNr were reduced,but GFAP increased gradually and GFAP filaments disapperared six months after MCAO.Conclusion Neuron reduction and gliocutes increase are the base of the atrophy of thalamus and SNr of distal areas after long-time MACO.%目的探讨持久性大脑中动脉闭塞(MCAO)后丘脑、黑质网状部(SNr)微管相关蛋白-2(MAP-2)及胶质纤维酸性蛋白(GFAP)的变化。方法 35只SD大鼠分为正常组、伪手术后2周组和1个月组、MCAO后2周组、1个月组、3个月组和6个月组等七组,每组5只。Tamura法制作动物模型。脑切片分别用鼠脑MAP-2、GFAP抗体,ABC法染色。观察丘脑的面积改变,SNr的面积和长度的改变;SNr神经细胞和胶质细胞数的变化。结果丘脑面积逐渐缩小;SNr面积长度及神经细胞减少,GFAP增加,6个月后丝极消失。结论神经细胞减少和胶质细胞增加是长期MCAO后丘脑和SNr萎缩的基础。
