目的 初步建立一种确定寄生菌被完全侵染的方法.方法 以细胞噬菌体T4(简称T4)侵染大肠杆菌为模型,通过3种方法测定不同滴度的T4在5 min内侵染模拟样品中大肠杆菌的效果.结果 在T4>1.00×108 PFU/mL的条件下侵染后,样液在培养中吸光度逐渐下降;在同样条件下侵染并清洗掉游离T4后,样液经平板培养无菌落产生,经液体培养不变混浊,并产生少量T4.结论 采用1.0×(108-109)PFU/mL的T4能够在5 min内有效而完全侵染模拟样品中大肠杆菌.%Objective To develop a method for detecting the fully infestation of bacterial parasites.Methods With bacteriophage T4 infested Escherichia coli(E.coli) as the experimental model,three tests were applied to detecting the effectiveness of different titer of T4 infesting E.coli in the analogue sample within 5 min.Results After 5 min infextation with T4 of more than 108 PFU/mL,the sample's OD600 decreased gradually during incubation.While,after infextation at the same condition,then washing away of free T4,the sample could not produce colonies of E.coli during incubation in LB agar plate.It could not become turbid during incubation in LB broth also,and produced a few T4.Conclusion T4 of 108 - 109 PFU/mL is essential and enough for infexting E.coli of the analogue sample efficiently and completely within 5 min.
