非小细胞肺癌中肺耐药蛋白的表达及与临床病理特征的关系

         

摘要

Objective To investigate the expression of lung resistance protein(LRP) in nonsmall-cell lung cancer(NSCLC)and its relationship with clinicopathologic characteristics. Methods Fluorescent quantitation-polymerases chain reaction(FQ-PCR) was used to detect the expression levels of LRP mRNA in 47 cases of NSCLC tissues and 14 cases of normal tissues. Results The positive rates of LRP in NSCLC and normal tissues were 74. 5%(35/47) and 14. 3%(2/14) respectively. The expression level was significantly higher in NSCLC tissuesCO. 58 + 0. 24)than that in normal issuesCO. 19 + 0. 17)0 = 5. 72,P<0. 001). The expression level of LRP was not correlated with age,sex,histological types,lymph node metastasis and clinical stage(P>0. 05) ,but was correlated with differentiation of cancer tissues(P = 0. 001). Conclusion There might be primary drug-resistance in NSCLC. Higher expression of LRP could play a major role in the intrinsic multi-drug resistance of NSCLC.%目的 探讨肺耐药蛋白(LRP)在非小细胞肺癌(NSCLC)中的表达及与临床病理特征的关系.方法 运用荧光定量-聚合酶链反应(FQ-PCR)检测47例肺癌组织和14例正常组织LRP mRNA的表达.结果 LRP mRNA在癌组织、正常组织中表达阳性率分别为74.5%(35/47)、14.3%(2/14),正常组织中LRP mRNA表达值为0.19±0.17,癌组织中为0.58±0.24,癌组织中LRP mRNA表达明显高于正常组织,差异有统计学意义(t=5.72,P=0.001);LRP mRNA表达与患者年龄、性别、组织学分型、淋巴转移及临床分期无关(P>0.05),而与肿瘤分化程度有关(P=0.001).结论 NSCLC中存在原发性耐药现象,LRP mRNA高表达在NSCLC对顺铂的原发性耐药中可能起重要作用.
