Objective To investigate the mechanism involved in the antitumor effects of Huaier .Methods The carboxyl fluores‐cein diacetate succinimide ester(CFSE) labeling method and Annexin V‐FITC/PI double‐staining were used to identify the effects of Huaier on A549 cells .The western blot was used to detect the protein levels of EZH2 ,β‐catenin and bcl‐2 after Huaier treatment in A549 cells .Results The CFSE labeling and Annexin V‐FITC/PI double‐staining results indicated that huaier suppressed prolifera‐tion and induced apoptosis of A549 cells by time and concentration dependent pattern .Western bloting result found that Huaier treatment down regulated EZH2 ,β‐catenin and bcl‐2 protein levels in A549 cells compared with untreated cells .Conclusion Huaier might suppress proliferation and induce apoptosis in lung cancer by EZH2‐mediated approach in lung cancer cells ,which provides a new idea for understanding the anti‐tumor effects of Huaier .%目的:探讨槐耳清膏抑制肺腺癌细胞增殖、促进细胞凋亡的抗肿瘤作用的分子机制。方法应用不同浓度槐耳清膏处理肺腺癌细胞,细胞增殖活性利用羧基荧光素二醋酸盐琥珀酰亚胺酯(CFSE)染色法检测,细胞凋亡应用 Annexin V‐FITC/PI双染流式细胞术检测,细胞中相关基因表达利用免疫技术检测。结果 CFSE 染色结果发现槐耳能够明显抑制肺腺癌细胞的增殖,并具有浓度和时间依赖性;Annexin V‐FITC/PI 双染流式细胞术检测细胞凋亡结果发现槐耳能够促进肺腺癌细胞凋亡,具有浓度和时间依赖性;免疫蛋白印迹实验结果发现在肺腺癌细胞中,槐耳能够抑制相关基因 EZH2、β‐catenin 和 bcl‐2的表达。结论在肺腺癌细胞中槐耳清膏抗肿瘤作用的分子机制可能是通过抑制 EZH2/β‐catenin 信号通路,抑制肺腺癌细胞增殖、促进细胞凋亡。
