非酒精性脂肪性肝炎大鼠肝组织过氧化物酶体增殖物激活受体αmRNA表达及清热化湿法对其影响的实验研究

         

摘要

Objectives: To study the effect of heat - clearing and damp - draining preparation on mRNA expression of peroxisome Prolifreceptor α ( PPARa) in hepatic tissue of rats with nonalcoholic steatohepatitis. Methods:The NASH models of rats were made by high fat diet. The rats were randomly divided into normal control, mode, formula of heat - clearing and damp - draining preparation and Compound methionine and choline control group. The content of serum triglyceride ( TG) , total cholesterol (TCH) , low density lipoprotein (LDL) , aminotransferase( ALT) and aspartate aminotransferase(AST) were detected with Olympus Au560 entire automated biochemical analysis instrument. mRNA expression of PPARain hepatic tissue of rats was detected by using in situ hybridization. Results: Compared with the normal control group, the PPARa mRNA expression of hepatic tissue of the blank model group was degraded obviously (P <0. 01 ). Compared with the blank model group, Formula of heat - clearing and damp - draining preparation and Compound Methionine and Choline Tablet could reinforce the PPARa mRNA expression of hepatic tissue (P <0. 05) , the difference of the effect between Formula of heat - clearing and damp - draining preparation group and Compound Methionine and Choline Tablet control group wasn't significant in statistics (P >0. 05 ). Conclusion; The PPARα mRNA expression of hepatic tissue degraded are nosogenesis of NASH. Formula of heat - clearing and damp -draining preparation can increase the PPARamRNA expression in hepatic tissue.%目的:观察NASH大鼠肝组织过氧化物酶体增殖物激活受体(PPARα) mRNA表达变化及加减王氏连朴饮对其影响.方法:采用高脂饮食复制大鼠NASH模型.分为正常对照组、空白模型组、加减王氏连朴饮组、东宝肝泰(复方蛋氨酸胆碱片)对照组.测定各组血清甘油三酯(TG)、总胆固醇(TCH)、低密度脂蛋白(LDL)和丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)水平;原位杂交技术检测肝组织PPARα mRNA的表达.结果:与正常对照组相比,空白模型组大鼠肝组织PPARα mRNA表达明显减弱(P<0.01);与空白模型组相比,加减王氏连朴饮组和东宝肝泰对照组大鼠肝组织PPARα mRNA表达明显增强(P<0.05),加减王氏连朴饮组和东宝肝泰对照组之间没有显著性差异(P>0.05).结论:PPARα mRNA表达减弱引起脂质代谢失衡参与NASH的发病.清热化湿方通过激活PPARα mRNA表达来调节脂质代谢,发挥防治NASH的作用.
