新城疫病毒TS09耐热株在BHK细胞上的传代培养

         

摘要

以新城疫病毒(Newcastle Disease Virus,NDV)TS09耐热株为研究对象,研究了该毒株在幼仓鼠肾传代细胞(BHK细胞)上的培养适应性.通过培养条件的优化、连续传代培养和极限稀释法纯化,获得了一株新的、纯化的、适应BHK细胞的NDV耐热株,命名为TS09-C株.TS09-C株的生物学特性测定结果表明,TS09-C株在BHK细胞中的增殖滴度为107.9 TCID50/mL,平均鸡胚致死时间(MDT)大于120 h,脑内致病指数(ICH)为0.与亲本株TS09株相比,TS09-C株的基因序列发生了较为明显的改变,但TS09-C株与亲本株仍保持着较近的遗传进化关系.%The characterization study of Newcastle disease virus (NDV) thermostable TS09 strain following serial passages in BHK cells was carried out. After optimization of the culture conditions, serial passages in BHK cells and purification by End-point dilutions, a newly purified, BHK cell-adapted NDV thermostable strain was developed, named TS09-C strain. Results from the biological characterization tests showed that the titer of TS09-C strain in BHK cells was 1079 TCID50/mL, MDT>120 h and ICPI was 0. It was demonstrated that there were a number of mutations in the genome of TS09-C strain, as compared with the TS09 strain. However, the close genetic relationships among the TS09-C and TS09 strains were observed.
